Cell Isolation Methods
Procedures for the isolation of rat and mouse aorta endothelial cells (AECs), aorta smooth muscle cells (ASMCs), pulmonary artery endothelial cells (PAECs), pulmonary artery smooth muscle cells (PASMCs), pulmonary microvascular endothelial cells (PMVECs), and pulmonary vein endothelial cells (PVECs).
Cell Characterization
Cell phenotypes are confirmed using a combination of antibodies, lectins, and acetylated LDL uptake. Endothelial cells are positive for factor VIII-Rag, eNOS, VE-cadherin, PECAM-1,and negative for smooth muscle cell alpha actin. The lectins Griffonia simplicifolia, Glycine max, and Helix pomatia are used to descriminate between endothelial cells derived from the various vascular segments.
